Johanna est \u00e9galement pr\u00e9sidente de la Soci\u00e9t\u00e9 finlandaise de biologie cellulaire et membre du conseil d'administration de l'Association finlandaise pour l'\u00e9tude de l'ob\u00e9sit\u00e9 (FASO : https:\/\/suomenlihavuustutkijat.fi\/<\/a>). Dans ses travaux r\u00e9cents, publi\u00e9s dans The Journal of Translational Medicine (disponibles \u00e0 l'adresse suivante : https:\/\/translational-medicine.biomedcentral.com\/articles\/10.1186\/s12967-024-05249-w<\/a>), Johanna et ses coll\u00e8gues ont \u00e9tudi\u00e9 les effets de l'ob\u00e9sit\u00e9 sur la s\u00e9cr\u00e9tion de v\u00e9sicules extracellulaires par les adipocytes humains et le tissu adipeux.<\/p>\n

\"A<\/p>\n

Les v\u00e9sicules extracellulaires (VE) constituent un groupe tr\u00e8s h\u00e9t\u00e9rog\u00e8ne de particules membranaires nanom\u00e9triques s\u00e9cr\u00e9t\u00e9es par tous les types de cellules mammif\u00e8res. Les VE sont pr\u00e9sentes dans tous les fluides corporels et transportent toutes sortes de biomol\u00e9cules pr\u00e9sentes dans les cellules \u00e0 partir desquelles elles sont s\u00e9cr\u00e9t\u00e9es. Ainsi, les VE sont bioactives, ce qui en fait d'importants communicateurs cellulaires dans l'hom\u00e9ostasie normale, mais aussi dans diverses maladies.<\/p>\n

Comme dans de nombreuses pathologies, des \u00e9tudes r\u00e9centes ont montr\u00e9 que les niveaux d'EV circulantes sont augment\u00e9s dans l'ob\u00e9sit\u00e9. Cependant, les recherches m\u00e9canistiques sur l'impact de l'ob\u00e9sit\u00e9 sur la s\u00e9cr\u00e9tion d'EV par les adipocytes et le tissu adipeux restent rares, et la plupart des \u00e9tudes pr\u00e9c\u00e9dentes ont utilis\u00e9 des EV provenant d'adipocytes et de tissu adipeux murins. De plus, les recherches pr\u00e9c\u00e9dentes se sont principalement concentr\u00e9es sur le contenu en prot\u00e9ines et en miARN des EV des adipocytes de souris, laissant le contenu en acides gras des EV largement sous-\u00e9tudi\u00e9. Matilainen et ses coauteurs ont donc \u00e9tudi\u00e9 le r\u00f4le des VE des adipocytes et du tissu adipeux dans l'ob\u00e9sit\u00e9 humaine, en utilisant une lign\u00e9e cellulaire d'adipocytes SGBS humains, du tissu adipeux visc\u00e9ral et sous-cutan\u00e9, ainsi que du plasma provenant de personnes ob\u00e8ses. Cette \u00e9tude a \u00e9galement fourni les premiers profils d\u00e9taill\u00e9s des acides gras des VE des adipocytes et du tissu adipeux humains.<\/p>\n

Les auteurs ont observ\u00e9 que les composants inflammatoires li\u00e9s \u00e0 l'ob\u00e9sit\u00e9 augmentent la s\u00e9cr\u00e9tion d'EV par les adipocytes humains et que le tissu adipeux visc\u00e9ral humain s\u00e9cr\u00e8te plus d'EV que le tissu adipeux sous-cutan\u00e9. \u00c0 la connaissance des auteurs, cette derni\u00e8re observation a \u00e9t\u00e9 d\u00e9montr\u00e9e pour la premi\u00e8re fois avec un soutien statistique. De plus, les VE plasmatiques des personnes ob\u00e8ses contenaient plus de VE positives \u00e0 l'adiponectine que celles des personnes de poids normal, ce qui confirme encore davantage la s\u00e9cr\u00e9tion plus importante de VE par le tissu adipeux en cas d'ob\u00e9sit\u00e9. Le nombre d'EV plasmatiques \u00e9tait en corr\u00e9lation positive avec l'IMC et l'expression du TNF dans le tissu adipeux sous-cutan\u00e9, \u00e9tablissant un lien entre l'augmentation de la s\u00e9cr\u00e9tion d'EV et l'expansion adipeuse et l'inflammation. Pour la premi\u00e8re fois gr\u00e2ce \u00e0 la microscopie confocale haute r\u00e9solution des EV, les auteurs ont fourni la preuve que les EV des adipocytes peuvent agir comme transporteurs de l'exc\u00e8s de lipides. Par rapport aux \u00e9tudes pr\u00e9c\u00e9dentes men\u00e9es sur des mod\u00e8les murins, des diff\u00e9rences ont \u00e9galement \u00e9t\u00e9 observ\u00e9es dans les sch\u00e9mas de s\u00e9cr\u00e9tion des EV et leurs profils d'acides gras.<\/p>\n

Ces r\u00e9sultats sugg\u00e8rent que les VE du tissu adipeux jouent un r\u00f4le important dans la r\u00e9ponse au stress et la redistribution des lipides dans l'ob\u00e9sit\u00e9 humaine. Cette \u00e9tude ouvrant de nombreuses possibilit\u00e9s, Johanna pr\u00e9voit de poursuivre ses recherches en tant que chercheuse postdoctorale afin d'explorer plus en profondeur les m\u00e9canismes m\u00e9di\u00e9s par les VE dans les pathologies li\u00e9es \u00e0 l'ob\u00e9sit\u00e9.<\/p>\n
